Sample preparation

The Nadia Instrument can encapsulate cells of over 50 μm due to its wide chip junction size. This includes cells that require different buffers such as cardiomyocytes and plant cells. Most instruments in the field can only process cells up to 30 µm and use standard sample buffers such as PBS.
Ease of use combined with accommodation of a wide range of cells and sizes.

Single cell library construction on Nadia

The RNAdia 2.0 kit enables the construction of single cell libraries on the Nadia Instrument, which consist of cellular transcripts, ready for sequencing. The Nadia Instrument is an automated, microfluidic droplet-based platform used for single cell research that encapsulates up to 8 samples, in parallel, in under 30 mins.
We now offer droplet visualization using our brand-new Droplet Analyzer Software.


Next Generation Sequencing

The pooled barcoded cDNA libraries using the RNAdia 2.0 kit are compatible with standard NGS short-read sequencing using our standard protocol, allowing the analysis of thousands of transcriptional profiles of single cells in parallel. Additionally, due to the presence of full length 3’ cDNA, the protocol is easily amenable to long-read sequencing on Pacific Biosciences or Oxford Nanopore sequencers.

Which reagents for the single cell sequencing workflow does the RNAdia 2.0 kit contain?
The RNAdia 2.0 kit contains reagents for all steps from sample encapsulation, cDNA library preparation, PCR amplification to NGS-library preparation.

When will the RNAdia 2.0 kit be available for order?
RNAdia 2.0 kits will be available for order from the 30th November.

How many samples can be processed with one RNAdia 2.0 kit?
We offer two versions of the RNAdia 2.0 kit, to process either 2 or 8 samples.

Is there a limit to cell size on the Nadia Instrument?
The Nadia Instrument can process cells of over 50µm and those with different buffer requirements such as cardiomyocytes and plant cells which are typically much larger than 50µm in diameter.

How do I analyze my single cell data?
Single cell transcriptomic data can be analyzed using open-source pipelines or with our in-house service for data analysis. You are able to purchase data analysis and visualization from Dolomite Bio as you can a kit.

What sample types are compatible with the Nadia Instrument?
The Nadia Instrument and its associated RNAdia 2.0 kit are compatible with a very wide range of samples types owing to its flexibility in sample buffers and broad cell size range. Species that have been tested internally and published range from mammals, fungi and plants and are sourced from primary tissue and model organisms.
