Why use agarose for your single cell projects?

  Agarose’s spherical scaffold structure and ability to allow for the diffusion of nutrients and gases make it an ideal system for naturalistic mammalian and non-mammalian live cell studies, given its many similarities to the native physiological environment of living cells. Single cells encapsulated in agarose can, therefore, be maintained for extended periods of time within these individual three-dimensional microenvironments and used in various research including single cell experiments.

We recognize that scientific terminology can be vague or confusing and we want to let you know exactly what our technology does!

The nadAROSE kit produces small spheres of hydrogels which contain cells. We call these cell-containing Scaffolds. They provide a structure to contain the cells and their products or a matrix through which they can propagate.

We differentiate scaffolds from spheres of aggregated or cultivated cells which are often termed Spheroids. Likewise, Organoids is term used to describe a collection of cells with recognisable organ characteristics.

Scaffold – small sphere of hydrogel that contains a cell(s)
Spheroid – spherical aggregation of cells (sometimes can mimic characteristics of tissue)
Organoid – a simple organ, produce in vitro, which demonstrates some recognisable organ characteristics.

The Nadia Instrument

The Nadia Instrument is an automated, microfluidic droplet-based platform for single cell research that encapsulates up to 8 samples, in parallel, in under 20 mins. Up to a million single cells can be captured per cartridge in a run.

The fully automated Nadia Instrument guides users through all relevant steps of the experiment via an easy-to-use touchscreen interface. Single cell-containing agarose scaffolds are constructed on Nadia using the nadAROSE kit. The Nadia is an automated, microfluidic droplet-based platform for single cell research that encapsulates up to 8 samples, in parallel, in under 20 mins. Up to one million single cells can be captured in a single run.

Cell release

The scaffolds produced with the nadAROSE kit can be later digested to release the cells captured inside them. If you want to subject them to a set of entirely different conditions or process them for particular analyses, you can.

Cells can be liberated from their scaffolds for further analyses.

Is the agarose provided in the kits in powder form?
The agarose aliquots are provided as a solid gel that is melted before use.

How many samples can I run in parallel?
You can run from 1 to 8 samples in parallel.

How many cells can you run?
Up to 5000 cells/ul.

How can you recover cells from the agarose scaffolds?
The use of a combination of β-agarase, β-agarase buffer, and DMEM can help dissolve scaffolds while maintaining cell viability. See application note for more details. Note: the reagents for digesting agarose scaffolds are not provided in the nadAROSE kit.

How long can you store scaffolds after recovery?
The agarose scaffolds can remain stable for up to a week; however, we recommend immediately processing the scaffolds due to cell viability.

What is the size of scaffolds produced?
Median size is 75 um.

Is the nadAROSE protocol suitable for 3D cell culture?
Agarose is not a suitable substrate for promoting mammalian cell growth, so it is not suitable for 3D cell culture. For a 3D cell culture kit, please refer to the nadia3D kit.
