Single cell RNA sequencing

Single cell RNA Sequencing (scRNA-Seq) is a tool that provides access to the transcriptomes of thousands of single cells – giving unprecedented insight into tissues at single cell resolution. This offers vital information about heterogeneity and cell-specific dynamics, which is key to understanding many diseases, immunity, development and more. This is in contrast to traditional techniques, that either allowed analysis of a few genes in thousands of individual cells (e.g., in situ hybridization), or the expression profile of thousands of genes, but only on homogenized samples.

Single nuclei RNA sequencing

Single nuclei RNA Sequencing (sNuc-Seq) is a technique which uses isolated nuclei instead of whole cells to profile gene expression in cells which are difficult to isolate. Using droplet microfluidic technology this enables users to profile thousands of nuclei at low cost, in a highly parallel method

How it works

SNuc-seq uses droplet microfluidics to encapsulate single nuclei with uniquely barcoded mRNA capture beads. Following downstream processing and sequencing, the transcriptomes of thousands of single nuclei can be analysed, and unique cell types identified.

Nuclei can usually be rapidly and easily isolated from frozen tissues, without the extended incubation and processing required for isolating single cells. The rapid isolation of nuclei from complex tissue will allow researchers to acquire transcriptomes that are relatively unperturbed by the isolation procedure.

FAQs

Is there a limit to biologicals size on the Nadia Instrument?

The Nadia Instrument can process biologicals of over 50µm and those with different buffer requirements such as cardiomyocytes and plant cells which are typically much larger than 50µm in diameter.

How many samples can I run in parallel?

You can run from 1 to 8 samples in parallel.

How many cells can you run?

Up to 5000 cells/ul.

What sample types are compatible with the Nadia Instrument?

The Nadia Instrument and its associated RNAdia 2.0 kit are compatible with a very wide range of samples types owing to its flexibility in sample buffers and broad cell size range. Species that have been tested internally and published range from mammals, fungi and plants and are sourced from primary tissue and model organisms.

How do I analyze my single cell data?

Single cell transcriptomic data can be analyzed using open-source pipelines or with our in-house service for data analysis. You are able to purchase data analysis and visualization from Do Bio as you can a kit.
